This model evaluates the antimicrobial efficacy of test articles for treatment of superficial skin infections. Test articles may be administered by oral, systemic or topical routes. Microbial counts are measured.

PROCEDURE SUMMARY
Groups of 5 female specific-pathogen-free ICR (CD-1) immunocompetent mice are used. The fur on the back is removed and the epidermal layer is disrupted. Infection is initiated by placing a droplet containing pathogen suspension on the wound area. The vehicle, the reference antibiotic, and test substances are administered starting at 2 h after infection. Doses may be delivered by IV, SC, PO, IM, or IP administration. At 24 hours after the first treatment, animals are humanely euthanized and tissue is aseptically harvested from each test animal. The collected tissues are homogenized and pathogen counts are determined by dilution plating technique.
CRITERIA FOR SIGNIFICANCE
A significant (p < 0.05) decrease in the bacterial counts of the treated animals compared to the vehicle control group or the baseline control group is considered a significant difference.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
