Epilepsy is a common neurological disorder characterized by seizures that are caused by episodic abnormal electrical activity in the brain. Animal models play an essential role in discovering and developing new antiepileptic drugs. Seizures are usually induced by an electrical stimulation or a convulsant agent in normal animals. In the model described below, the maximal electroshock (MES) is used to characterize anticonvulsant properties of test compounds in rodents.

PROCEDURE SUMMARY
Test substance is administered orally (PO) to a group of 5 ICR male mice weighing 23 ± 3 grams (g) one hour before maximal electroshock (60 Hz sine wave, 50 mA, 200 msec duration) is applied through corneal electrodes. Appearance of maximal electroshock-induced tonic convulsions (MES) in less than 50% of mice indicates significant anticonvulsant activity. Suggested Testing: n=5/group (study design dependent). Anticonvulsant activity assessed at initial dose of 100 mg/kg (p.o.). Dosing volume at 10 mL/kg.

We will readily accommodate client-specified alterations. These studies are performed at our AAALAC accredited BSL-2 laboratory in Taipei, Taiwan. All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
