This model evaluates the antimicrobial efficacy of test articles for treatment of Pseudomonas aeruginosa PAO1 wound infection with immune-competent mice. Microbial counts are measured. Strain PAO1 is a commonly used and virulent laboratory strain.

PROCEDURE SUMMARY
Groups of 5 female specific-pathogen-free ICR (CD-1) mice weighing 22 ± 2 g are used. The fur on the back is removed by an electric clipper, and then a sharp punch (ID 12 mm) is used to remove the skin including the panniculus carnosus and adherent tissues while the animal remains anesthetized by isoflurane gas (3%-5%) at 2 hours before infection. Infection is initiated by placing a 5 μL suspension of Pseudomonas aeruginosa (1 x 10⁵ CFU), on the wound area. Vehicle and/or test substances are then administered at 1 and 13 hours later per the specification of the study design table. The animals are euthanized by CO₂ asphyxiation at specified time points after treatment initiation (24 hours to 3 days). The wound is excised andhomogenized with 1 mL of PBS. Homogenates( 0.1 mL) aliquots, are used for 10-fold serial dilutions and plated on Nutrient Broth in 1.5% Bacto Agar for bacterial counts.
CRITERIA FOR SIGNIFICANCE
A significant (p < 0.05) decrease in the bacterial counts of the treated animals compared to the vehicle control group or the baseline control group was considered a significant difference.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
