This model assesses the efficacy of test articles for protection against a lethal peritoneal infection with cytotoxic clinical isolate of Pseudomonas aeruginosa. It may be used for evaluating small molecules, vaccines and biologics.

PROCEDURE SUMMARY
Groups of 10 ICR (CD-1) derived female mice weighing 22 ± 2 gms are used. Each animal is inoculated with an intraperitoneally administered lethal dose of Pseudomonas aeruginosa (PA6206) suspended in 0.5 ml brain-heart infusion broth containing 5% mucin. Test substance and vehicle are administered 1 and 13 hours later (or per the client request) by IP, PO, IV or SC injection. The dose (up to 100 mg/kg) and vehicle (recommended 2% Tween 80 or 0.9% NaCl, 10 ml/kg) are specified by the sponsor. Mortality is recorded daily during the following 7 days. Prevention of mortality in 50 percent or more (≥ 50%) of the animals indicates significant activity.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
