BACKGROUND INFORMATION
This model assesses the efficacy of test articles for protection against a lethal lung infection with cytotoxic clinical isolate of Pseudomonas aeruginosa in immune-competent mice. It may be used to assess efficacy of small molecules, vaccines and biologics.

PROCEDURE SUMMARY
Groups of 10 BALB/c derived female mice weighing 18 ± 2 g are used. On day 0, animals are inoculated intranasally (0.02 mL/lung) of Pseudomonas aeruginosa aeruginosa (PA6206) after anesthesia with etomidate (~20 mg/kg, IV) or pentobarbital (40 mg/kg, IV). Vehicle and/or test substances are then administered (IV, PO, IP, or SC) 1 and 13 hours later (or per the client request). Mortality is recorded daily during the following 10 days. Prevention of mortality in 50 percent or more (≥ 50%) of the animals indicates significant activity.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
