BACKGROUND INFORMATION
This model assesses the antimicrobial and protective efficacy of test articles in a lethal lung tissue infection with a carbapenem resistant P. aeruginosa clinical isolate. The model is conducted with immune-competent mice and may therefore be useful for evaluating immune modulatory agents, antibody therapeutics, and vaccines in addition to direct-acting small molecule antibacterial agents.

PROCEDURE SUMMARY
Groups of 10 BALB/c derived female mice weighing 18 ± 2 g are used. Each animal is inoculated intranasally with a LD90-100 dose (2-4 x 10⁷ CFU/mouse) of Pseudomonas aeruginosa (AR-Bank#0266). Test substance and vehicle are administered one hour later by IP, PO, IV, or SC injection. Following test substance treatment as above, mortality is recorded daily during the following 10 days. Mortality data is graphed with the Kaplan Meier Method, and Significance is assessed with the Fisher's exact test. The mean survival time of vehicle and test article treatment groups are compared with ANOVA.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
