BACKGROUND INFORMATION
This assay is used to assess the antimicrobial efficacy of test articles in a murine model of complicated urinary tract infection model using neutropenic mice. Pathogen is injected directly into the kidney tissue, antimicrobial agent is administered then the change in pathogen load is measured following a 24 h treatment period. The pyelonephritis model has been established with diverse Enterobacteriaceae including susceptible and multidrug resistant strains of E. coli, K. pneumoniae, E. cloacae, and P. mirabilis to enable testing of organisms with varied antibiotic susceptibility and resistance mechanisms.

PROCEDURE SUMMARY
Groups of 5 female specific-pathogen-free ICR (CD-1) neutropenic mice are directly inoculated with pathogen suspension into kidneys. Vehicle and/or test substances are then administered by IV, IP, SC, or PO administration (or per client's request) starting at 2 hours after infection. At 24 hours after the first treatment, animals are humanely euthanized and both kidneys are harvested from each test animal. The collected kidneys are homogenized and pathogen counts are determined by dilution plating technique.
CRITERIA FOR SIGNIFICANCE
A significant (p < 0.05) decrease in the bacterial counts of the treated animals compared to the vehicle control group or the baseline control group is considered a significant difference.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
