PROCEDURE SUMMARY
Groups of 10 male or female specific-pathogen-free BALB/c weighing 18 ± 2 g are used. Each immunocompetent (IC) animal is inoculated intranasally (0.02 mL/mouse) with an LD90-100 dose (1-3 x 10^5 CFU) of Klebsiella pneumoniae (ATCC 43816) suspended in PBS. Vehicle and/or test substances are then administered 1 and 13 hours later (BID) or per client’s specifications. Standard dose routes are IV, IP, SC or PO. Mortality is recorded daily during the following 7 days. Prevention of mortality is recorded in percentage and the significance is assessed by Fisher’s exact test. For bacterial enumeration (optional service), the animals are euthanized with CO2 asphyxiation at 25 h post-infection, and the lung tissue is harvested from each of the test animals. The removed lung tissues are homogenized in 1 mL PBS, pH 7.4, with a Polytron homogenizer. Homogenates, 0.1 mL, are diluted 10-fold and plated on MacConkey II agar for colony count (CFU/g) determination.

We will readily accommodate client-specified alterations. These studies are performed at our AAALAC accredited BSL-2 laboratory in Taipei, Taiwan. All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
