Stress-induced abnormal defecation using a rat model of conditioned fear stress (CFS), which is known to be psychological stress model useful for the evaluation of anxiolytic, antidepressant, and anti-IBS agents.

PROCEDURE SUMMARY
Groups of 6 male Sprague-Dawley (SD) rats weighing 200 ± 20 g are used. Animals are trained twice at 24-hours interval in an electric-shock machine with electrode grid-inlaid floor with a program of 3 sec warning sound followed with 5 sec electric shock and intensive light per minute as a trial with a total of 30 exposures per day. Forty-eight (48) hours later, rats are returned to the electric shock chamber to evoke conditioned fear stress (CFS) with warning sound and light in the absence of electric shock for 30 trials to collect the CFS induced defecation. Vehicle, test articles and loperamide are administered orally (PO) 1 hour before the CFS challenge. The incidence of diarrhoea is noted. The severity of diarrhoea is assessed by using an arbitrary scoring scale ranging from 0 to 2: normal stools (0), soft stools (1) and liquid stools (2). In addition, the weight of stool is also measured in this study.
