PROCEDURE SUMMARY
Groups of 10 BALB/c female mice weighing 18 ± 1 g are used. Each animal is inoculated with a lethal dose (1 x 10⁸ CFU/mouse) of Escherichia coli (ATCC 25922) suspended in PBS by intravenous injection. Test substance and vehicle are administered 1 hour and 13 hours after inoculation or per the client's specifications. Standard dose routes are IP, IV, or SC injection. Mortality is recorded twice daily during the following 10 days. Prevention of mortality is recorded and compared to the vehicle treatment group. The significance is assessed with the Fisher's exact test. Satellite infected and treated animals, 5 mice per group, may be included in the study for bacterial enumeration from blood and tissues. Animals are sacrificed at 8 hours after infection.

We will readily accommodate client-specified alterations. These studies are performed at our AAALAC accredited BSL-2 laboratory in Taipei, Taiwan. All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
