The 5-HT2A agonist 4-Iodo-2,5-dimethoxosyamphtetamine (DOI) is employed as a prototype to access the relative activity of selected serotonin agonists by measuring the number of head twitches in mice.

PROCEDURE SUMMARY
Groups of 8 C57BL/6 male mice, at 7 ± 1 weeks old, are employeed. Test substance is administered orally 1 hour before testing. The mice are dosed with DOI at 3 mg/kg through SC. The mice are immediately placed into individual beakers (1000 mL). Each mouse is individually observed following the injection of DOI, and the head-twitch responses (HTR) counts are recorded cumulatively at 5-minute intervals for the next 20 minutes through visual observation (mean ± SEM). One-way ANOVA followed by Dunnett’s test is applied for a comparison between vehicle and treated groups. Differences are considered significant at p<0.05.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
