BACKGROUND INFORMATION
The vitamin D3 analog MC903 (calcipotriol), a psoriasis therapeutic drug, is used to induce atopic dermatitis (eczema-like symptoms), including Th2 type inflammation and eosinophilia. The MC903-induced atopic dermatitis (eczema) mouse model is commonly used to study the mechanisms of skin inflammation and to evaluate therapeutic efficacy.

PROCEDURE SUMMARY
Groups of 8 female C57BL/6 mice at 8 weeks old are used. MC903 (2 nmole/20 µL, dissolved in 100% ethanol) is topically (TOP) applied daily on the right ear one hour after treatment from Day 1 to Day 14 for 14 consecutive days. Control mice receive the same volume of ethanol during the same study period. Vehicle, test articles and reference compound, tacrolimus ointment (0. 02 mg/ear), are administered topically (TOP) once daily from Day 1 to Day 14 for 14 consecutive days (1 hour before MC903 challenge). Body weight is recorded daily during the study. Ear swelling is measured by an experimenter blinded to treatment group, with a dial thickness micrometer gauge as an index of inflammation 30 min before dosing on Days 1, 3, 5, 7, 9, 11, and 13. On Day 15, ear swelling is measured 24 hours after the last dosing. The animals are sacrificed and then the ears are harvested and weighed. Ear edema is calculated by subtracting the thickness of Day 1 (normal control) from Day n (treated ear). Percentage inhibition is calculated according to the formula: (Ic −It)/Ic x 100, where Ic and It refers to the increase of ear thickness (mm) both in the control and treated animals. Additional endpoint measurements can be requested include clinical scoring, histopathology, MPO, cytokines, and FACS etc.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
