BACKGROUND INFORMATION
Anxiety is a complex combination of emotions that includes various components: fear, apprehension and worry (cognitive) often accompanied by sensations such as palpitations, nausea, chest pain and/or shortness of breath (somatic) as well as panic (emotional) and escape and/or avoidance of the perceived source of anxiety (behavioral).  Anxiolytics may, therefore, be useful in the treatment of various disorders including Generalized Anxiety Disorder (GAD), Panic Disorder, Simple Phobia, Social Anxiety Disorder, Obsessive Compulsive Disorder, Posttraumatic Stress Disorder and Substance-Induced Anxiety Disorder.  Agents that relieve anxiety, termed anxiolytics or minor tranquillizers, may be therapeutically useful in the treatment of the above disorders.

PROCEDURE SUMMARY
The elevated plus-maze is shaped like a plus sign with two unwalled (open) arms (50 cm x 10 cm) and two walled (closed) arms (50 cm x 10 cm x 40 cm). The floor of each arm and the walls of the closed arms are constructed of black plexiglass. The entire maze is elevated to a height of 50 cm. Test substance is administered orally (PO) to a group of 6 Sprague-Dawley (SD) male rats weighing 130 ± 20 g. After one hour, each test animal is placed at the center of the plus-maze facing one of the open arms. During a five-minute test period, the time that each animal spends in the open arms is recorded. The mean ± SEM of the time spent in open arms for each treatment group is calculated. One-way ANOVA followed by Dunnett’s test is applied for a comparison between vehicle and treated groups. Differences are considered significant at p<0.05. Suggested Testing: n=6/group (study design dependent). Anti-anxiety effects assessed at an initial dose of 30 mg/kg (p.o.). Dosing volume at 10 mL/kg.

Time spent in open arms of plus-maze is caculatedand. ANOVA followed by Dunnett's test is applied for comparison between test compound and vehicle control groups.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
