BACKGROUND INFORMATION
The Minimum Inhibitory Concentration (MIC) Assay assesses the in vitro antimicrobial potency of the test article(s). In this assay, the MIC is the lowest concentration of an agent that completely inhibits visible growth. The MIC protocol with Neisseria gonorrhoeae follows the Clinical and Laboratory Standards Institute (CLSI) agar dilution method. A panel of N. gonorrhoeae strains was tested (FA1090 ATCC 700825, AR Bank #0179-StrR, AR Bank #0181-StrR, and H041 NCTC 13820-StrR).

PROCEDURE SUMMARY
The agar dilution method follows the guidelines of the Clinical and Laboratory Standards Institute (CLSI), M07-A11, and thus yields data that is suitable for publication or regulatory submission. The antimicrobial test article is incorporated into an agar medium at eight concentrations varying by two-fold serial dilutions. Medium for N. gonorrhoeae is GC agar base with 1% defined growth supplement. Pathogen suspension, 0.2 µl, is spotted on the test medium surface with a replicator device yielding 1-2 x 10⁴ CFU/spot. The inoculated agar plates are incubated at 36 ± 1°C in an atmosphere of containing 5% CO₂ for 20 to 24 hours before reading the MICs. Complete, 100% visual growth inhibition is the MIC endpoint criteria. Vehicle control and an active antibiotic were used as blank and positive controls. Alterations to the protocol can be accommodated with advanced notification.
