BACKGROUND INFORMATION
Thioacetamide (TAA) is widely used to induce acute toxic liver injury, and specially causes membrane damage, oxidative stress and lipid proxidation. Thioacetamide-induced hepatic injury is commonly used to study the mechanisms of hepatic inflammation and evaluate the therapeutic efficacy.

PROCEDURE SUMMARY
Groups of 6 male C57BL/6 mice at 7-8 weeks of age are used. Animals are fasted overnight prior to use. Test article is administered by oral gavage (PO) at 1 hour before thioacetamide (TAA, 400 mg/kg, IP) challenge. The animals are sacrificed 24 hours after the administration of thioacetamide, and blood is collected. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels are measured by automated biochemical analyzer TBA-120FR. Additional analyses that can be requested include ALP, T-BIL and ALB levels, liver weight, biomarker analysis (protein and/or mRNA) and histopathology etc. Doses may be administered PO, IV, IP and SC. Suggested Testing: n=6/group (study design dependent).

We will readily accommodate client-specified alterations. These studies are performed at our AAALAC accredited BSL-2 laboratory in Taipei, Taiwan. All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011). The study protocol was approved by the Pharmacology Discovery Services IACUC and is performed with the oversight of veterinarians to assure the humane treatment of laboratory animals.
