BACKGROUND INFORMATION
Bile Duct Ligation (BDL) is widely accepted and used to induce liver cholestasis and fibrosis. In the bile duct ligation protocol, rats are subjected to double ligation of the common bile duct with dissection of the bile duct between the ligatures. This model is applied to investigate the anti-inflammatory and anti-fibrotic effects of test articles for hepatic inflammation and fibrosis in rats.

PROCEDURE SUMMARY
Groups of 8 male SD rats weighing 220 ± 20 g are used. Rat are anesthetized by inhalation of 1.5~3.0 Vol% isoflurane and the surgical area is shaved and swabbed with 75% alcohol and iodine, and then laparotomy is performed. The exposed bile duct is ligated with 4-0 silk and secured with two double ligatures around the proximal and distal parts of the bile duct. The bile duct is divided by the scissors. The abdomen is closed in two layers with 4-0 silk suture and swabbed with iodine. In the sham group, the abdomen is closed without BDL surgery. Vehicle and test articles are administered orally at 2 hours after BDL surgery (Day 0) and once daily for 2 or 4 weeks. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels are measured on Day 7 and 14 or Days 14 and 28 by optimized UV method with an automatic analyzer (TBA-120 FR, Toshiba, Japan). Body weight is recorded three times a week. The animals are sacrificed 24 hours after the final treatment (Day 14 or Day 28) and the liver is harvested and weighed. Liver-to-body weight ratio is then calculated for each animal according to the formula: Liver (g)/BW x 100.
