BACKGROUND INFORMATION
Allergic contact dermatitis (ACD), which is caused by allergen, is a type of delayed hypersensitivity. Contact hypersensitivity (CHS) is a T-cell mediated immune response in the epidermis. The fluorescein isothiocyanate (FITC)-induced CHS model in mice is commonly used to study the mechanisms of skin inflammation and evaluate the therapeutic efficacy.

PROCEDURE SUMMARY
Groups of 8 male BALB/c mice at 8 weeks of age are used. Two hundred microliter (200 µL) of 0.5% Fluorescein Isothiocyanate (FITC) dissolved in 20% tributyrin in acetone is applied topically (TOP) onto the shaved abdominal skin on Days 1 and 2, respectively. On Day 7, mice are challenged by applying 20 µL of 0.5% FITC solution onto both sides of the right ear.
Vehicle, test articles and reference compound are administered topically (TOP) or orally (PO) once daily from Day 6 to Day 9 for 4 consecutive days (1 hour before FITC challenge on Day 7). Body weight is recorded daily during the study period. Ear swelling is measured by an experimenter blinded to treatment group, with a dial thickness micrometer gauge as an index of inflammation 30 min before dosing on Days 7, 8, 9 and 10. On Day 10, ear swelling is measured at 24 hours after the last dosing. The animals are sacrificed and the ears are then harvested and weighed. Ear edema is calculated by subtracting the thickness of Day 7 (normal control) from Day n (treated ear). Percentage inhibition is calculated according to the formula: (Ic – It)/Ic x 100, where Ic and It refers to the increase of ear thickness (mm) both in the control and treated animals. Additional endpoint measurements can be requested, including clinical scoring, histopathology, MPO, cytokines, and FACS etc.

All aspects of this work are performed in general accordance with the Guide for the Care and Use of laboratory animals (National Academy Press, Washington, DC, 2011).
